B lymphocyte-specific, Cre-mediated mutagenesis in mice.

چکیده
Adaptation of the P1 phage-derived Cre /loxP site- specific recombination system to the gene targeting technique allows for the conditional deletion of genes in mice. To selectively modify genes in B lymphocytes, we have generated mice (designated CD19-Cre) which express cre under the transcriptional control of the B lineage-restricted CD19 gene. In a model system involving the cross of CD19-Cre mice with mice bearing a loxP -flanked substrate, we find a deletion efficiency of 75-80% in bone marrow-derived pre-B cells that increases to 90-95% in splenic B cells.

Kinetics of Primary and Memory Cytotoxic T Lymphocyte Responses to Herpes Simplex Virus 1 Infection: Granzyme B Mediated CTL Activity
Background: Herpes simplex virus type 1 is one of the most common viruses among human population. Studies demonstrate the essential role of cell mediated immunity, especially CD8+ T cells, in prevention and clearance of HSV1. Objective: It is of great importance to improve our knowledge about the kinetics of CTL responses to primary and secondary HSV-1 infection. Methods: Using a sensitive tech...
متن کاملSite-specific somatic mutagenesis in the retinal pigment epithelium.
PURPOSE Generate site-specific somatic mutations selectively in the retinal pigment epithelium (RPE) in mice. METHODS A transgenic mouse line expressing the Cre recombinase under the control of the tyrosinase-related protein (TRP)-1 promoter was generated. The presence of Cre was determined by in situ hybridization, and Cre-mediated excision of DNA was analyzed by PCR and alkaline phosphatase...
متن کاملExpression of the SNARE Protein SNAP-23 Is Essential for Cell Survival
Members of the SNARE-family of proteins are known to be key regulators of the membrane-membrane fusion events required for intracellular membrane traffic. The ubiquitously expressed SNARE protein SNAP-23 regulates a wide variety of exocytosis events and is essential for mouse development. Germline deletion of SNAP-23 results in early embryonic lethality in mice, and for this reason we now descr...
متن کاملTemporally and spatially regulated somatic mutagenesis in mice.
In mice transgenesis through oocyte injection or DNA recombination in embryonal stem (ES) cells allows mutations to be introduced into the germline. However, the earliest phenotype of the introduced mutation can eclipse later effects. We show in mice that site-specific genomic recombination can be induced in a selected cell type, B lymphocytes, at a chosen time. This precision of somatic mutage...
متن کاملSpatio-temporally controlled site-specific somatic mutagenesis in the mouse.
The efficient introduction of somatic mutations in a given gene, at a given time, in a specific cell type will facilitate studies of gene function and the generation of animal models for human diseases. We have shown previously that conditional recombination-excision between two loxP sites can be achieved in mice by using the Cre recombinase fused to a mutated ligand binding domain of the human...
